Endurance exercise induces various adaptations that yield health benefits; however, the underlying molecular mechanism has not been fully elucidated. Given that it has recently been accepted that inflammatory responses are required for a specific muscle adaptation after exercise, this study investigated whether toll-like receptor (TLR) 4, a pattern recognition receptor that induces proinflammatory cytokines, is responsible for exercise-induced adaptations in mouse skeletal muscle. The TLR4 mutant (TLR4m) and intact TLR4 control mice were each divided into 2 groups (sedentary and voluntary wheel running) and were housed for six weeks. Next, we removed the plantaris muscle and evaluated the expression of cytokines and muscle regulators. Exercise increased cytokine expression in the controls, whereas a smaller increase was observed in the TLR4m mice. Mitochondrial markers and mitochondrial biogenesis inducers, including peroxisome proliferator-activated receptor beta and heat shock protein 72, were increased in the exercised controls, whereas this upregulation was attenuated in the TLR4m mice. In contrast, exercise increased the expression of molecules such as peroxisome proliferator-activated receptor-gamma coactivator 1-alpha and glucose transporter 4 in both the controls and TLR4m mice. Our findings indicate that exercise adaptations such as mitochondrial biogenesis are mediated via TLR4, and that TLR4-mediated inflammatory responses could be involved in the mechanism of adaptation.

The nasal strip is widely used in horses during exercise, but effects of using a nasal strip are controversial and little is known about its effect on horses undergoing endurance events. The aim of this study was to determine whether the use of nasal strips influences alveolar cell population assessed by bronchoalveolar lavage (BAL), tidal volume, and nasal airflow rate. Six Arabian horses were subjected to two low intensity tests on a treadmill, with and without application of a commercial external nasal strip. Tidal volumes and airflow rates were measured during the test; two hours after the test, BAL was performed to assess cytology of pulmonary secretions. The lavage fluid showed increased neutrophil count after exercise in animals with the nasal strip (P<0.05). This suggests that turbulence of airflow through the nasal cavity may have diminished with nasal strip use, thus allowing larger particles to be deposited more distally in the respiratory system, inducing a more intense neutrophilic response. No differences in tidal volumes or airflow rates were observed between groups (with or without nasal strips) during the test (P>0.05). The use of nasal strips seems to influence alveolar cell population during and after exercise in horses after low intensity exercise tests. Further studies are needed to verify whether alveolar cell population is related to poor exercise performance in horses.(AU)


A fita nasal é amplamente utilizada em equinos durante o exercício, porém seus resultados são controversos e pouco conhecidos em animais que disputam provas de resistência. O objetivo deste trabalho foi verificar se o uso da fita nasal influencia a população de células alveolares por meio de lavado broncoalveolar, volume corrente e fluxo de ar nasal. Foram utilizados seis equinos da raça Árabe, que realizaram dois testes de longa duração em esteira, sendo um teste com a fita e outro sem a fita nasal. Fluxo e volume respiratório foram mensurados durante o teste; duas horas após o término do exercício, foi realizado lavado broncoalveolar para realização de citologia da secreção pulmonar. Foi verificada maior porcentagem de neutrófilos após o exercício nos animais que se exercitaram com a fita nasal (P<0,05), indicando que o turbilhonamento na passagem do ar através da cavidade nasal pode ter diminuído, permitindo que partículas maiores se depositassem em porções mais distais do sistema respiratório, induzindo uma resposta neutrofílica mais intensa. Não houve diferenças entre os parâmetros ventilatórios analisados entre os animais quando correram com ou sem a fita nasal (P>0,05). O uso da fita nasal parece influenciar alguns parâmetros, durante e após o exercício, em animais que realizam provas de longa duração. Outros estudos devem ser realizados para verificar se essa influência pode melhorar o desempenho desses animais em exercícios predominantemente aeróbicos.(AU)

Serotonin is a neurotransmitter synthesized by the amino acid tryptophan, that has the potential to impact the behaviour and activity of dogs. The objective of this study was to assess the effects of supplemental tryptophan and a 12-week incremental training regimen on the voluntary activity and behaviour of client-owned Siberian Huskies. Sixteen dogs were blocked for age, BW and sex and then randomly allocated to either the control or treatment group. Both groups were fed the same dry extruded diet; however, the treatment group were supplemented with tryptophan to achieve a tryptophan: large neutral amino acid ratio of 0.075:1. Once a week, a 5-minute video recording was taken immediately pre- and post- exercise to evaluate dogs' behaviours. Activity monitors were used to record voluntary activity on both training and rest days. Linear regression analysis was used to assess the relationship between training week and time spent performing each behaviour. Additionally, a repeated measure mixed model was used to test differences between diet groups and training week for both behavioural and activity count data. The time spent performing agonistic behaviours prior to exercise was negatively associated with week for treatment dogs (ß = -0.32, 95% CI [-0.55, -0.10], P < 0.05) and no change was observed for control dogs (ß = -0.13, 95% CI [-0.41, 0.15], P > 0.10). Treatment did not have any effect on activity levels (P > 0.10). For all dogs, locomotive behaviours decreased prior to exercise as weeks progressed (P < 0.05), while run day voluntary activity depended on the distance run that day (P < 0.05). These data suggest that sled dogs experience an exercise-induced reduction in voluntary locomotion in response to both single bouts and repetitive bouts of exercise. Additionally, tryptophan supplementation may decrease agonistic behaviours, without having any effect on voluntary activity.
